Module 5 - The EU-SOL Resource and Technology Platform

This Module addresses the establishment of resources for tomato and potato gene expression profiling. This will be achieved through further development of oligonucleotide-based microarrays presently under development or available commercially. At the start of EU-SOL the best opportunities for selection of a platform will be discussed. Transcription factor profiling by real-time PCR will be set up, as well as a TILLING and ecoTILLING platform to target genes related to tomato quality traits. The French tomato TILLING initiative will be integrated within a European context.  For functional analysis of genes in tomato and potato, a transactivation system using two-components and a system for virus-induced gene silencing (VIGS) will be established, while vectors for efficient RNAi silencing will be constructed. These systems and vectors will be established centrally in a few labs and the resources and technologies will be distributed to other participants to facilitate their research in the modules. In order to maintain a common genetic framework that is in line with the strategic objectives of EU-SOL we have chosen a limited set of genes, some of which will be targeted by the various platforms during the first 18 months of the project (Core-Gene). This limited set of genes will allow us to enter the second phase of the project with preliminary results that will guide future directions. Core-Gene modified genotypes obtained by various approaches will be used by the various profiling platforms (metabolome, transcriptome, consumer preference) and set the ground for future evaluation of yet, uncharacterized genes and biological phenomena.
Module5
Linked to the International SOL Genome Sequencing Programme EU-SOL will allocate resources for tomato genome sequencing. Duplication of other international efforts (USA, India, China, Korea, and Japan) will be avoided by exchange of materials and data. This investment will strengthen the cooperation with SOL genome initiatives in INCO countries such as Argentina and Brazil as well as global cooperation of tomato researchers.
